nhr-25::yfp is expressed exclusively (but ubiquitously) in hypodermis/epidermis. First fluorescence appears approximately 150-190 minutes after fertilization (20°C).
Variegation of intensity in the dorsal hyp6 and hyp7 precursors gives reasonable visualization of cell fusion events. Strong cytoplasmic staining in ventral hyp precursors reveals protrusive extensions coincident with migration to the ventral midline. Engulfed cell corpses are visible as dark figures within fluorescent cytoplasm of phagocytic hyp cells.
Movies made with hanging-embryo mounts (random DV/RL orientation for each embryo).
Integrated transgene array.
All four movies have the embryo misoriented with anterior to the right.
